研究概要	本総合研究Aの平成3年度の研究成果の概要は以下のとおりである。 1.イエバエのビテロゲニンmRNAレベルの変動をRNAブロツテイング法にて解析した。雌では脂肪体中のビテロゲニンmRNA量の変動は体液中ビテロゲニンと平行関係にあった。雌雄の遊離腹部を用いた実験から、脱皮ホルモンがビテロゲニン合成を誘導することが判明した。カイコ・ビテロゲニン・サブユニットのNー末端の一次構造とcDNAの塩基配列の比較から、H鎖とし鎖は一本のmRNAより翻訳生成されることを確認した。カイコ卵特異的タンパク質の全塩基配列を決定,推定アミノ酸配列は哺乳類リパ-ゼと高い相同性を示した。カイコSP1受容体タンパク質と脂肪体細胞膜上に同定した。 2.カイコ・フェノ-ルオキシダ-ゼの活性化に関与するタンパク質を分離精製し,治性化カスケ-ドを再構築後,反応機構の解析を行った。カイコ・外皮ラッカ-ゼおよびショウジョウバエ・プロフェノ-ルオキシダ-ゼのアイソザイムを分離精製した。ショウジョウバエではプロパノ-ルによってもフェノ-ルオキシダ-ゼが活性化されることを確認。 3.ニクバエ・アリルホリン遺伝子5´上流域の塩基配列ACCACAACAに特異的に結合するタンパク質を幼虫脂肪体より精製した。このタンパク質は40Kと42Kの2種のサブユニットより構成され、ニクバエ胚由来の培養細胞核より調製した無細胞転写系でアリルホリン遺伝子の正確な転写を促進した。 4.トノサマバッタにおける脂質動員ホルモン(AKH)と長距離飛翔との関連性について解析した。AKHの分泌により血中トレハロ-スが減少し、トレハロ-スによるジグリセリドの積み込みが解除され,脂肪体から飛翔筋への大量の脂質輸送が開始されることが判明した。
